Le sonde d’ibridazione LC , altrimanti chiamate sonde FRET probes, sono marcate con un singolo quencher fluorescente. Due sonde d’ibridazione LC specifiche per due sequenze adiacenti nel DNA target sono utilizzate per Real-Time PCR quantitativa.

Le sonde LC sono generalmente sintetizzate con FAM come fluoroforo donatore al 3’, e  ROX o Cy® 5 come fluoroforo accettore al 5’. Viene aggiunto un  gruppo 3’ fosfato  per prevenire l’estensione della sonda reporter da parte della Taq DNA polymerase durante i cicli di PCR. Tutte le sonde sono purificate HPLC purified e controllate tramite MALDI-TOF Mass Spectrometry.

Storage conditions: -20 °C to -70 °C. Oligonucleotides are stable in solution at 4°C for up to 2 weeks. Properly reconstituted material stored at -20°C should be stable for at least 6 months. Lyophilized DNA (when kept at -20°C) in a nuclease-free environment should be stable for years.
